We focus on understanding the cellular and molecular biological mechanisms underlying neurological diseases, with a specific emphasis on Sigma-1R, a versatile chaperone protein implicated in numerous neurodegenerative conditions. Our ongoing laboratory projects encompass two main areas: (I) exploring the molecular mechanisms and functions of Sigma-1R in the nuclear pore and nuclear envelope in neurodegenerative diseases, and (II) developing a screening platform for Sigma-1R agonists or antagonists with translational applications. Ultimately, we posit that Sigma-1R could serve as a promising pharmacological target for mitigating neurological diseases. In addition, our research delves into C9orf72 amyotrophic lateral sclerosis (ALS)/frontotemporal dementia (FTD), a prominent genetic contributor to familial ALS, as well as its presence in sporadic cases. Specifically, we investigate dipeptide repeat protein (DPR), particularly the arginine-rich DPR, which increases motor neuron death and promotes neuroinflammation. The project outlines three primary objectives: I) examining whether Sigma-1R can safeguard against motor neuron death in C9orf72 ALS/FTD by promoting nucleocytoplasmic transport function, II) assessing whether Sigma-1R can alleviate DPR-induced neuroinflammation by inhibiting glial activation, and III) establishing in vitro and in vivo models for DPR-induced C9orf72 ALS/FTD.
